Anit-SPam settings in Control Panel

I have anti-spam setup on my email accounts through the control panel. I have an email account which forwards from my ISP account to my account hosted with Jodo, my ISP's server has been blacklisted (good thing I don't send with that account) and the anti-spam controls are blocking my forwarded messages.

If I remove the anti-spam from my email account will it let that email through or do all black listed servers get stopped regardless of the spam settings?

Once changes are made to the spam settings how long do they take to become effective?
 
Messages from Blacklisted servers are not accepted irrespective of the anti-spam settings in control panel.
