This error has nothing to do with an app pool, it is a 404 meaning it can’t find the file.

This problem looks like a portal alias. The dnn will pickup any alias and point it to an installs home page. If you try to navigate elsewhere, you will normally get an issue because the comp you are using doesn’t have localhost/whatever installed. Go in and add the domain alias and not just localhost/whatever
If you fails to do so then install the latest security patches from Microsoft like the Security Update for Microsoft .NET Framework, Version 1.1 Service Pack 1 (KB886903)? After installation reboot the server.
